Title: Fabrication of Fibrin Scaffold for Corneal Regeneration

Purpose:

Damage to the cornea, caused by infection, trauma, and mechanical disorders, may lead to loss of corneal transparency and functionality, which cause partial or complete loss of vision among other complications. One of the most common methods for corneal replacement is the surgical transplantation of the cornea, which is confined by the shortage of suitable donor tissue. In these cases, the risk of disease transfer, high cost, and suture-related complications have shed light on the importance of an alternative biomaterial. Recently, fibrin-based scaffolds revealed great potential in ocular tissue regeneration as well as tissue regeneration of other parts of the body.

Methods:

In this study, the fibrin gel has been synthesized by optimizing the concentration of fibrinogen and thrombin to improve the mechanical properties, degradation time and cell response of fibrin-like scaffolds.

Results:

Mechanical analysis of optimized fibrin gel showed the Young module and tensile strength of up to 72 and 121 kPa, respectively. Additionally, the results demonstrated that the degradation time of fibrin gel was enhanced up to 7 days in the presence of aprotinin which was much longer time compared to that of fibrin gel without a protease inhibitor. Human corneal fibroblasts (HCFs) were seeded on fibrin gel for 5 days. The fibrin gel showed excellent cell viability over 5 days, and the proliferation of HCFs also increased significantly after 3 days.

Conclusion:

The fibrin gel illustrates the great potential to use in which appropriate stability, mechanical properties, and cell growth are essential to tissue functionality and regeneration.
